Unconventional detection methods for microfluidic devices 总被引:2,自引:0,他引:2
The direction of modern analytical techniques is to push for lower detection limits, improved selectivity and sensitivity, faster analysis time, higher throughput, and more inexpensive analysis systems with ever-decreasing sample volumes. These very ambitious goals are exacerbated by the need to reduce the overall size of the device and the instrumentation - the quest for functional micrototal analysis systems epitomizes this. Microfluidic devices fabricated in glass, and more recently, in a variety of polymers, brings us a step closer to being able to achieve these stringent goals and to realize the economical fabrication of sophisticated instrumentation. However, this places a significant burden on the detection systems associated with microchip-based analysis systems. There is a need for a universal detector that can efficiently detect sample analytes in real time and with minimal sample manipulation steps, such as lengthy labeling protocols. This review highlights the advances in uncommon or less frequently used detection methods associated with microfluidic devices. As a result, the three most common methods - LIF, electrochemical, and mass spectrometric techniques - are omitted in order to focus on the more esoteric detection methods reported in the literature over the last 2 years. 相似文献

A film of niobium oxide was immobilized on a SiO2/C carbon-ceramic matrix (specific surface area 270 m2 g−1) and characterized by N2 adsorption-desorption isotherms, scanning electron microscopy, X-ray photoelectron spectroscopy and atomic force microscopy. This new carbon-ceramic material, SiO2/C/Nb2O5, was used for construction of electrodes, and it shows ability to improve the electron-transfer between the electrode surface and ascorbic acid. The electrocatalytic oxidation of ascorbic acid was made by differential pulse and cyclic voltammetry techniques, making it potentially useful for developing a new ascorbic acid sensor. 相似文献

On-chip pressure injection for integration of infrared-mediated DNA amplification with electrophoretic separation 总被引:1,自引:0,他引:1
Poly(dimethylsiloxane) (PDMS) membrane valves were utilized for diaphragm pumping on a PDMS-glass hybrid microdevice in order to couple infrared-mediated DNA amplification with electrophoretic separation of the products in a single device. Specific amplification products created during non-contact, infrared (IR) mediated polymerase chain reaction (PCR) were injected via chip-based diaphragm pumping into an electrophoretic separation channel. Channel dimensions were designed for injection plug shaping via preferential flow paths, which aided in minimizing the plug widths. Unbiased injection of sample could be achieved in as little as 190 ms, decreasing the time required with electrokinetic injection by two orders of magnitude. Additionally, sample stacking was promoted using laminar or biased-laminar loading to co-inject either water or low ionic strength DNA marker solution along with the PCR-amplified sample. Complete baseline resolution (Res = 2.11) of the 80- and 102-bp fragments of pUC-18 DNA marker solution was achieved, with partially resolved 257- and 267-bp fragments (Res = 0.56), in a separation channel having an effective length of only 3.0 cm. This resolution was deemed adequate for many PCR amplicon separations, with the added advantage of short separation time-typically complete in <120 s. Decreasing the amount of glass surrounding the PCR chamber reduced the DNA amplification time, yielding a further enhancement in analysis speed, with heating and cooling rates as high as 13.4 and -6.4 degrees C s(-1), respectively. With the time requirements greatly reduced for each step, it was possible to seamlessly couple IR-mediated amplification, sample injection, and separation/detection of a 278-bp fragment from the invA gene of <1000 starting copies of Salmonella typhimurium DNA in approximately 12 min on a single device, representing the fastest PCR-ME integration achieved to date. 相似文献

This study highlights the potential utility of high-performance capillary electrophoresis (HPCE) for monitoring enzyme activity. Free-zone capillary electrophoresis is used to rapidly and reproducibly analyze the activity of the bacterial enzyme chloramphenicol acetyl transferase (CAT) which converts the substrates acetyl coenzyme A (CoA) and chloramphenicol to acetyl chloramphenicol and CoA. The results of this study indicate that HPCE may be an excellent tool for studying enzyme activities since it has several advantages over standard single parameters assays, most notably, the ability to monitor both loss of substrate and appearance of products simultaneously. Conditions have been identified for optimal separation of the substrate (chloramphenicol) from the products (acetylated derivatives). This presents a unique potential of HPCE for the analysis of enzymatic reactions that may be applied to areas of analytical research presently utilizing enzymatic reactions. One such analytical method is the CAT assay used for analysis of gene promoter activity. In this study, HPCE is shown to yield similar quantitative results with nonradiolabelled substrate in a fraction of the time. HPCE has several advantages over standard techniques including speed of analysis, no need for radiolabelled substrate, small sample volumes, high sensitivity/resolution and excellent quantitative capabilities. 相似文献

Matrix-assisted laser desorption/ionization (MALDI) mass spectra were obtained from single biological aerosol particles using an aerosol time-of-flight mass spectrometer (ATOFMS). The inlet to the ATOFMS was coupled with an evaporation/condensation flow cell that allowed the aerosol to be coated with matrix material as the sampled stream entered the spectrometer. Mass spectra were generated from aerosol composed either of gramicidin-S or erythromycin, two small biological molecules, or from aerosolised spores of Bacillus subtilis var niger. Three different matrices were used: 3-nitrobenzyl alcohol, picolinic acid and sinapinic acid. A spectrum of gramicidin-S was generated from approximately 250 attomoles of material using a molar ratio of 3-nitrobenzyl alcohol to analyte of approximately 20:1. A single peak, located at 1224 Da, was obtained from the bacterial spores. The washing liquid and extract solution from the spores were analyzed using electrospray mass spectrometry and subsequent MS/MS product ion experiments. This independent analysis suggests that the measured species represents part of the B. subtilis peptidoglycan. The on-line addition of matrix allows quasi-real-time chemical analysis of individual, aerodynamically sized particles, with an overall system residence time of less than 5 seconds. The potential for using polyelectrolyte multilayers (PEMs) to provide chromatographic functionality on continuous silica networks created from sol-gel chemistry has been evaluated by capillary electrochromatography (CEC). Construction of the PEM was achieved by flushing the column with polyelectrolytes of alternative charge, with variation of the properties of the exposed polyelectrolyte providing a unique means to vary the chromatographic surface. Variation of the exposed polyelectrolyte from poly(diallyldimethylammonium chloride) (PDDAC) to dextran sulfate (DS) allowed the direction of the electroosmotic flow (EOF) to be changed and also provided a means to vary the chromatographic capacity. Variation of negative polymer from DS to poly(styrene sulfonate) (PSS) significantly altered the EOF and the migration of peptides, with both the reversed-phase and ion-exchange capacities increasing. An alternative method for changing the column capacity was to change the thickness of the PEM, which was evaluated by anion-exchange CEC. A 70-80% increase in retention was observed for all anions without any increase in EOF suggesting significant penetration of the analytes through the PEM and interaction with buried charges within the PEM. 相似文献

Single- and double-electron transfer to autoionizing 1s2l2l(') configurations in fluorine ions have been investigated for 1.1 MeV/u collisions of F7+ and F8+ with He and Ne. The resulting Auger electron emission spectra show anomolously large intensities for the formation of the metastable 1s(2s2p 3P) 4P state compared to the similarly configured 1s(2s2p 3P) 2P- and 1s(2s2p 1P) 2P+ states. The large 4P intensity, which cannot be explained on the basis of spin statistics, is attributed instead to the Pauli exchange of similarly aligned electrons. 相似文献

We have used complete correlated momentum mapping of the photoelectron and heavy ion products from the dissociation of the di-cation of acetylene, induced by photoionizing the carbon K shell of one of the atoms, to map out the angular correlation between the electron and the axis of the target molecule. The (quasi-) symmetric decay is found to proceed through both acetylene and vinylidene configurations. By using the strongly peaked photoelectron emission to "start a clock," an upper limit of 60 fs is placed on the isomerization time from the acetylene to the vinylidene configuration. 相似文献

Hollow-state (double K-shell vacancy) production in Li-like Be+, B2+, C3+, and F6+ ions colliding with He has been investigated using high-resolution Auger-electron spectroscopy. The formation of discrete states and their relative intensities show striking dependences on the nuclear charge Z of the ion. From the projectile velocity dependences of these states the contribution of the electron-electron interaction is determined, showing that the hollow states are formed largely by electron correlation with a strength that diminishes for increasing Z. 相似文献
